AKTIVITAS PENURUNAN KADAR KOLESTEROL TOTAL EKSTRAK DAUN BANGKAL (NAUCELIA SUBDITA) SECARA IN VITRO

Abstrak

Hiperkolesterolemia menyebabkan terjadinya peningkatan lemak dalam darah sehingga salah satunya menyebabkan peningkatan kadar Kolesterol.Hiperkolesterolemia menyebabkan terjadinya pelemakan di sel hepar berupa vakuola lemak diikuti oleh adanya vakuola berisi air hingga terjadi nekrosis bahkan sirosis, hal tersebut dapat mengganggu fungsi hepar sebagai tempat metabolisme karbohidrat, lemak, dan protein. Penelitian ini merupakan penelitian eksperimental dengan bertujuan untuk mengetahui aktivitas ekstrak daun bangkal (Nauclea subdita) dalam menurunkan kadar kolesterol secara in vitro. Metode Analisis aktivitas penurunan kolesterol dilakukan dengan metode Lieberman Burchard pereaksi asam sulfat dan asetat anhidrat ,dan metode TFC dilakukan dengan perekasi Alcl3 Ekstrak daun bangkal mengandung senyawa flavonoid, alkaloid, fenolik, terpenoid, saponin, dan tannin. Hasil TLC didapatkan nilai Rf sampel dengan baku pembanding selisihnya sedikit / hampir sama jadi didaptkan posotif (+) flavonoid 0.724selesain itu warna bercak yang dihasilkan bewarna biru berpijar , steroid didaptkan hasil rata -rata 0.024 dimana mendekati dengan nilai baku pembanding 0.023. hasil TFC ekstrak daun bangkal (Nauculea subdita) rata -rata 1184.633±120.3155 mgQE/g , penurunan kadar kolestrol total Ec₅₀ sebesar 97.448 ± 56.10824ppm . Ec₅₀ sebesar 27.354ppm untuk simvastatin Berdasarkan hasil penelitian dan pembahasan penelitian maka dapat diperoleh kesimpulan bahwa ektrak daun bangkal (Nauculea subdita) lebih besar dari simvastatin sebagai baku pembanding esktrak daun bangkal dapat menurunkan kadar kolestrol97.448 ± 56.10824ppm.

This study is an experimental research with the aim of determining the activity of bangkal leaf extract (Nauclea subdita) in reducing cholesterol levels in vitro. Method The analysis of cholesterol-lowering activity was carried out by the Lieberman Burchard method of reacting sulfuric acid and anhydrous acetate, and the TFC method was carried out by Alcl3 pereking Bangkal leaf extract contains flavonoid compounds, alkaloids, phenolics, terpenoids, saponins, and tannins. The results of TLC were obtained with the Rf value of the sample with the benchmark standard, the difference was slight / almost the same, so the positive (+) flavonoid 0.724 was obtained, the color of the spots produced was incandescent blue, steroids were obtained, the average result was 0.024 which was close to the benchmark value of 0.023. TFC results of Bangkal leaf extract (Nauculea subdita) averaged 1184,633±120,3155 mgQE/g, decrease in total cholesterol levels of Ec₅₀ by 97,448 ± 56.10824ppm. Ec₅₀ of 27,354ppm for simvastatin Based on the results of the study and research discussion, it can be concluded that bangkal leaf extract (Nauculea subdita) is greater than simvastatin as a benchmark for comparison of brackish leaf extract can reduce cholesterol levels97,448 ± 56,10824ppm.
